肿瘤细胞增殖是生物体的重要生命特征,细胞以分裂的方式进行增殖。单细胞生物,以细胞分裂的方式产生新的个体。多细胞生物,以细胞分裂的方式产生新的细胞,用来补充体内衰老和死亡的细胞;同时,多细胞生物可以由一个受精卵,经过细胞的分裂和分化,zui终发育成一个新的多细胞个体。必须强调指出,通过细胞分裂,可以将复制的遗传物质,平均地分配到两个子细胞中去。可见,肿瘤细胞增殖是生物体生长、发育、繁殖和遗传的基础。真核细胞的分裂方式有三种,即有丝分裂、无丝分裂和减数分裂。
活细胞线粒体中的琥珀酸脱氢酶能使外源性MTT还原为水不溶性的蓝紫色结晶甲瓒(Formazan)并沉积在细胞中,而死细胞无此功能。然后采用二甲基亚砜(DMSO)溶解细胞中的甲瓒,用酶联免疫检测仪在570 nm波长处测定其光吸收值,反映活细胞数量。在一定细胞数范围内,MTT结晶形成的量与肿瘤细胞数成正比。
